Principle of the Assay: P21 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-P21 antibody and an P21-HRP conjugate. The assay sample and buffer are incubated together with P21-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the P21 concentration since P21 from samples and P21-HRP conjugate compete for the anti-P21 antibody binding site. Since the number of sites is limited, as more sites are occupied by P21 from the sample, fewer sites are left to bind P21-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The P21 concentration in each sample is interpolated from this standard curve.

This gene encodes a potent cyclin-dependent kinase inhibitor. The encoded protein binds to and inhibits the activity of cyclin-CDK2 or -CDK4 complexes, and thus functions as a regulator of cell cycle progression at G1. The expression of this gene is tightly controlled by the tumor suppressor protein p53, through which this protein mediates the p53-dependent cell cycle G1 phase arrest in response to a variety of stress stimuli. This protein can interact with proliferating cell nuclear antigen (PCNA), a DNA polymerase accessory factor, and plays a regulatory role in S phase DNA replication and DNA damage repair. This protein was reported to be specifically cleaved by CASP3-like caspases, which thus leads to a dramatic activation of CDK2, and may be instrumental in the execution of apoptosis following caspase activation.
