Principle of the assay: AMHR2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AMHR2 antibody and an AMHR2-HRP conjugate. The assay sample and buffer are incubated together with AMHR2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AMHR2 concentration since AMHR2 from samples and AMHR2-HRP conjugate compete for the anti-AMHR2 antibody binding site. Since the number of sites is limited, as more sites are occupied by AMHR2 from the sample, fewer sites are left to bind AMHR2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AMHR2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the receptor for the anti-Mullerian hormone (AMH) which, in addition to testosterone, results in male sex differentiation. AMH and testosterone are produced in the testes by different cells and have different effects. Testosterone promotes the development of male genitalia while the binding of AMH to the encoded receptor prevents the development of the mullerian ducts into uterus and Fallopian tubes. Mutations in this gene are associated with persistent Mullerian duct syndrome type II. Alternatively spliced transcript variants encoding different isoforms have been identified.
