Principle of the Assay: AMTN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AMTN antibody and an AMTN-HRP conjugate. The assay sample and buffer are incubated together with AMTN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AMTN concentration since AMTN from samples and AMTN-HRP conjugate compete for the anti-AMTN antibody binding site. Since the number of sites is limited, as more sites are occupied by AMTN from the sample, fewer sites are left to bind AMTN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AMTN concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The mineralized portions of teeth, the dentin and enamel, are formed by mesenchyme-derived odontoblasts and epithelium-derived ameloblasts, respectively. As ameloblasts differentiate, they deposit specific proteins necessary for enamel formation, including amelogenin (AMELX; MIM 300391), enamelin (ENAM; MIM 606585), and ameloblastin (AMBN; MIM 601259), in the organic enamel matrix. Amelotin is specifically expressed in maturation-stage ameloblasts (Iwasaki et al., 2005 [PubMed 16304441]).[supplied by OMIM, Mar 2008]

Product Notes
The Human AMTN amtn (Catalog #AAA7233819) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7233819 ELISA Kit recognizes Human AMTN. It is sometimes possible for the material contained within the vial of "Amelotin (AMTN), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
