Intended Uses: The A1M test kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring A1M in human biological samples. If the ELISA is to be used outside the intended use, the user may need to optimize for said use.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the Alpha 1-Microglobulin (A1M) present in samples reacts with the anti-A1M antibodies, which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-A1M antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound A1M. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of A1M in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of A1M in the test sample. The quantity of A1M in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

NCBI Description

This gene encodes a complex glycoprotein secreted in plasma. The precursor is proteolytically processed into distinct functioning proteins: alpha-1-microglobulin, which belongs to the superfamily of lipocalin transport proteins and may play a role in the regulation of inflammatory processes, and bikunin, which is a urinary trypsin inhibitor belonging to the superfamily of Kunitz-type protease inhibitors and plays an important role in many physiological and pathological processes. This gene is located on chromosome 9 in a cluster of lipocalin genes. [provided by RefSeq]

1. increased protein levels in preeclamptic women
2. significant relationship was established between the low level and absence of bikunin, and pro-u-PA in urine samples from patients with bladder tumors; identified bikunin loss in urine as a potential bladder carcinoma marker
3. Overexpression of Urinary trypsin inhibitor is associated with recurrence in hepatocellular carcinoma.
4. The transfer of heavy chains from bikunin proteins to hyaluronan requires both TSG-6 and HC2.
5. Overexpression of bikunin reduced the gene expression of matriptase, which attenuated in vitro cell invasion in prostate cancer.

9. AMBP has radical scavenging activity.
10. Bikunin was found to localize on the cell membrane, while tryptase was in the secretary granules of the mast cells from psoriatic lesions.
11. Free uristatin and bikunin pass readily into urine and are primarily bound to heavy chains that constitute the proinhibitor form in plasma.
12. small angle X-ray scanning study of the monomer and dimer forms of alpha1-microglobulin; monomer is characterised by a radius of gyration R(G)= 2.20 nm and D(max)=6.3 nm and the dimer by R(G)=2.99 nm and D(max)=9.5 nm

Product Notes

The Human AMBP ambp (Catalog #AAA564034) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA564034 ELISA Kit recognizes Human AMBP. It is sometimes possible for the material contained within the vial of "alpha 1-Microglobulin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
