Principle of the assay: CRK ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CRK antibody and an CRK-HRP conjugate. The assay sample and buffer are incubated together with CRK-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRK concentration since CRK from samples and CRK-HRP conjugate compete for the anti-CRK antibody binding site. Since the number of sites is limited, as more sites are occupied by CRK from the sample, fewer sites are left to bind CRK-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRK concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of an adapter protein family that binds to several tyrosine-phosphorylated proteins. The product of this gene has several SH2 and SH3 domains (src-homology domains) and is involved in several signaling pathways, recruiting cytoplasmic proteins in the vicinity of tyrosine kinase through SH2-phosphotyrosine interaction. The N-terminal SH2 domain of this protein functions as a positive regulator of transformation whereas the C-terminal SH3 domain functions as a negative regulator of transformation. Two alternative transcripts encoding different isoforms with distinct biological activity have been described. [provided by RefSeq, Jul 2008]

Phosphorylation induces a change in intramolecular folding and SH2 interactions, causing its rapid dissociation from the tyrosine kinase complex.
