Human ADAM10 (A Disintegrin And Metalloprotease 10) ELISA Kit
Principle of the Assay: This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human ACV-A. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human ACV-A and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human ACV-A, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Human ACV-A. You can calculate the concentration of Human ACV-A in the samples by comparing the OD of the samples to the standard curve!!Background/Introduction: Activin A is a disulphide-linked homodimer of the b-subunit of inhibin, a member of the transforming growth factor-b (TGF-b) 1 superfamily of proteins [1]. Although four activin genes have been discovered in humans, most studies have focused on the cell biology of activin A. This molecule is involved in the regulation of mitogenesis, embryogenesis, apoptosis, wound repair and other regulatory processes in vivo. Activin A has also been shown to play important roles in the pluripotency, proliferation and differentiation of embryonic stem cells, underpinning an emerging area of biomedical research. Activin A is initially produced as a large precursor molecule, with the pro-region separated from the mature region by a polybasic motif [1]. This motif is putatively cleaved in mammalian cells by subtilisin-like pro-protein convertases (SPCs), such as furin to produce the mature protein. Activin A has been purified from several sources, including porcine follicular fluid and ovine amniotic fluid [2].

Also cleaves the ectodomain of the integral membrane proteins CORIN and ITM2B. May regulate the EFNA5-EPHA3 signaling.
