Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Kallikrein 7 (KLK7) were tested 20 times on one plate, respectively.
Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Kallikrein 7 (KLK7) were tested on 3 different plates, 8 replicates in each plate.
CV(%) = SD/meanX100
Intra-Assay: CV<10%
Inter-Assay: CV<12%
Test Principle
The microplate provided in this kit has been pre-coated with an antibody specific to Kallikrein 7 (KLK7). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Kallikrein 7 (KLK7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Kallikrein 7 (KLK7) level in the sample or standard.
Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

This gene encodes a member of the kallikrein subfamily of serine proteases. These enzymes have diverse physiological functions and many kallikrein genes are biomarkers for cancer. The encoded protein has chymotrypsin-like activity and plays a role in the proteolysis of intercellular cohesive structures that precedes desquamation, the shedding of the outermost layer of the epidermis. The encoded protein may play a role in cancer invasion and metastasis, and increased expression of this gene is associated with unfavorable prognosis and progression of several types of cancer. Polymorphisms in this gene may play a role in the development of atopic dermatitis. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene, which is one of fifteen kallikrein subfamily members located in a gene cluster on chromosome 19. [provided by RefSeq, May 2011]

Uniprot Description

KLK7: May catalyze the degradation of intercellular cohesive structures in the cornified layer of the skin in the continuous shedding of cells from the skin surface. Specific for amino acid residues with aromatic side chains in the P1 position. SCCE cleaves insulin B chain at '6-Leu-|-Cys-7', '16-Tyr-|-Leu-17', '25-Phe-|-Tyr-26' and '26-Tyr-|-Thr-27'. Could play a role in the activation of precursors to inflammatory cytokines. Belongs to the peptidase S1 family. Kallikrein subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 3.4.21.35; Protease; Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 19q13.41

Cellular Component: extracellular region

Molecular Function: serine-type peptidase activity; serine-type endopeptidase activity

Biological Process: extracellular matrix disassembly; extracellular matrix organization and biogenesis; epidermis development; proteolysis

Product Notes

The Human KLK7 klk7 (Catalog #AAA2708997) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2708997 CLIA Kit recognizes Human KLK7. AAA Biotech's Kallikrein 7 (KLK7) can be used in a range of immunoassay formats including, but not limited to, Chemiluminescence Immunoassay (CLIA). Researchers should empirically determine the suitability of the KLK7 klk7 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Kallikrein 7 (KLK7), CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
