• Test Principle: The microplate provided in this kit has been pre-coated with an antibody specific to Glycine Dehydrogenase (GLDC). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Glycine Dehydrogenase (GLDC). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Glycine Dehydrogenase (GLDC) level in the sample or standard.
• Preparation and Storage: The stability of kit is determined by the loss rate of activity. Degradation of glycine is brought about by the glycine cleavage system, which is composed of four mitochondrial protein components: P protein (a pyridoxal phosphate-dependent glycine decarboxylase), H protein (a lipoic acid-containing protein), T protein (a tetrahydrofolate-requiring enzyme), and L protein (a lipoamide dehydrogenase). The protein encoded by this gene is the P protein, which binds to glycine and enables the methylamine group from glycine to be transferred to the T protein. Defects in this gene are a cause of nonketotic hyperglycinemia (NKH).[provided by RefSeq, Jan 2010]
