(Jurkat cells were grown in a T175 flask at 37 degree C, 5% CO2, in DMEM with 10% FBS. Cells were centrifuged and resuspended in 4 mL of DMEM without serum at 20x106 cells/mL. Cells were then stimulated with 30 mM H2O2(4 mL of 2X) for 15 min. After the incubation, cells were lysed by adding 2 mL of 5X Lysis Buffer #1 containing 5 mM NaF and 10 mM Na3VO4, on an orbital shaker (400 rpm) for 30 min at room temperature. The media was removed from the wells, and cells were lysed with 20 uL/well of 5X Lysis Buffer #1 containing 5 mM NaF and 10 mM Na3VO4, on an orbital shaker (400 rpm) for 30 min at room temperature. The lysates were then analyzed for total-SLP76 using the two-plate assay protocol. Lysates analyzed on separate wells for phospho-SLP76 (S376) are also shown for comparison. The TR-FRET signal was recorded at 665 and 615 nm (EnVision lamp excitation) after an 18 h incubation period at room temperature.)

This gene encodes an adapter protein that acts as a substrate of the T cell antigen receptor (TCR)-activated protein tyrosine kinase pathway. The encoded protein associates with growth factor receptor bound protein 2, and is thought to play a role TCR-mediated intracellular signal transduction. A similar protein in mouse plays a role in normal T-cell development and activation.
