Background: Prostate-specific antigen, also known as Gamma-seminoprotein, Kallikrein-3, kallikrein-related peptidase 3, Semenogelase, KLK3 and PSA, is a secreted protein which belongs to thepeptidase S1 family and Kallikrein subfamily. KLK3/Kallikrein 3 contains one peptidase S1 domain. KLK3/Kallikrein 3 is a glycoprotein produced almost exclusively by the prostate gland. KLK3/Kallikrein 3 is produced for the ejaculate where it liquifies the semen in the seminal coagulum and allows sperm to swim freely.It is also believed to be instrumental in dissolving the cervical mucous, allowing the entry of sperm. Human KLK3/Kallikrein 3 and human KLK2 are closely related products of the human kallikrein genes KLK3 and KLK2, respectively. Both KLK3/Kallikrein 3 and human kallikrein 2 are produced and secreted in the prostate and have important applications in the identification of prostate cancer. NCBI Description

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates several transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

PSA: Hydrolyzes semenogelin-1 thus leading to the liquefaction of the seminal coagulum. Forms heterodimer with SERPINA5. Inhibited by SERPINA5. Activity is strongly inhibited by Zn2+, 100 times more abundant in semen than in serum. This inhibition is relieved by exposure to semenogelins, which are avid zinc binders.
