Principle of the assay: HMGB1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HMGB1 antibody and an HMGB1-HRP conjugate. The assay sample and buffer are incubated together with HMGB1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HMGB1 concentration since HMGB1 from samples and HMGB1-HRP conjugate compete for the anti-HMGB1 antibody binding site. Since the number of sites is limited, as more sites are occupied by HMGB1 from the sample, fewer sites are left to bind HMGB1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HMGB1 concentration in each sample is interpolated from this standard curve.

Uniprot Description

HMGB1: DNA binding proteins that associates with chromatin and has the ability to bend DNA. Binds preferentially single-stranded DNA. Involved in V(D)J recombination by acting as a cofactor of the RAG complex. Acts by stimulating cleavage and RAG protein binding at the 23 bp spacer of conserved recombination signal sequences (RSS). Heparin-binding protein that has a role in the extension of neurite-type cytoplasmic processes in developing cells. Component of the RAG complex composed of core components RAG1 and RAG2, and associated component HMGB1 or HMGB2. Belongs to the HMGB family.

Product Notes

The Sheep HMGB1 hmgb1 (Catalog #AAA741573) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA741573 ELISA Kit recognizes Sheep HMGB1. It is sometimes possible for the material contained within the vial of "High Mobility Group box1 Protein, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
