Human Hexokinase 2 ELISA Kit
Principle of the Assay: HK2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HK2 antibody and an HK2-HRP conjugate. The assay sample and buffer are incubated together with HK2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HK2 concentration since HK2 from samples and HK2-HRP conjugate compete for the anti-HK2 antibody binding site. Since the number of sites is limited, as more sites are occupied by HK2 from the sample, fewer sites are left to bind HK2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HK2 concentration in each sample is interpolated from this standard curve.

Hexokinases phosphorylate glucose to produce glucose-6-phosphate, the first step in most glucose metabolism pathways. This gene encodes hexokinase 2, the predominant form found in skeletal muscle. It localizes to the outer membrane of mitochondria. Expression of this gene is insulin-responsive, and studies in rat suggest that it is involved in the increased rate of glycolysis seen in rapidly growing cancer cells. [provided by RefSeq]

Polymorphism: Although found in NIDDM patients, genetic variations of HK2 do not contribute to the disease.
Miscellaneous: In vertebrates there are four major glucose-phosphorylating isoenzymes, designated hexokinase I, II, III and IV (glucokinase).
