Specificity
This assay has high sensitivity and excellent specificity for detection of HE4. No significant cross-reactivity or interference between HE4 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between HE4 and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 pmol/ml

Intended Uses: This HE4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse HE4. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: HE4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HE4 antibody and an HE4-HRP conjugate. The assay sample and buffer are incubated together with HE4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HE4 concentration since HE4 from samples and HE4-HRP conjugate compete for the anti-HE4 antibody binding site. Since the number of sites is limited, as more sites are occupied by HE4 from the sample, fewer sites are left to bind HE4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HE4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a protein that is a member of the WFDC domain family. The WFDC domain, or WAP Signature motif, contains eight cysteines forming four disulfide bonds at the core of the protein, and functions as a protease inhibitor in many family members. This gene is expressed in pulmonary epithelial cells, and was also found to be expressed in some ovarian cancers. The encoded protein is a small secretory protein, which may be involved in sperm maturation. [provided by RefSeq, Jul 2008]

Product Notes

The Mouse HE-4 wfdc2 (Catalog #AAA752662) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA752662 ELISA Kit recognizes Mouse HE-4. It is sometimes possible for the material contained within the vial of "Epididymis Protein 4, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
