Principle of the Assay: This assay employs the direct competitive inhibition enzyme immunoassay technique. Antibody specific for HB-beta has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with biotin-conjugated HB-beta. A competitive inhibition reaction is launched between HB-beta (Standards or samples) and biotin-conjugated HB-beta with the pre-coated HB-beta antibody. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in opposite to the amount of HB-beta bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The alpha (HBA) and beta (HBB) loci determine the structure of the 2 types of polypeptide chains in adult hemoglobin, Hb A. The normal adult hemoglobin tetramer consists of two alpha chains and two beta chains. Mutant beta globin causes sickle cell anemia. Absence of beta chain causes beta-zero-thalassemia. Reduced amounts of detectable beta globin causes beta-plus-thalassemia. The order of the genes in the beta-globin cluster is 5'-epsilon -- gamma-G -- gamma-A -- delta -- beta--3'.
