Principle of the Assay: GSTT2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GSTT2 antibody and an GSTT2-HRP conjugate. The assay sample and buffer are incubated together with GSTT2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GSTT2 concentration since GSTT2 from samples and GSTT2-HRP conjugate compete for the anti-GSTT2 antibody binding site. Since the number of sites is limited, as more sites are occupied by GSTT2 from the sample, fewer sites are left to bind GSTT2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GSTT2 concentration in each sample is interpolated from this standard curve.

NCBI Description

Cytosolic and membrane-bound forms of glutathione S-transferase are encoded by two distinct supergene families. These enzymes function in the detoxification of electrophilic compounds, including carcinogens, therapeutic drugs, environmental toxins and products of oxidative stress, by conjugation with glutathione. The genes encoding these enzymes are known to be highly polymorphic. These genetic variations can change an individual's susceptibility to carcinogens and toxins as well as affect the toxicity and efficacy of some drugs. At present, eight distinct classes of the soluble cytoplasmic mammalian glutathione S-transferases have been identified: alpha, kappa, mu, omega, pi, sigma, theta and zeta. This gene encodes a glutathione S-tranferase belonging to the alpha class. The alpha class genes, located in a cluster mapped to chromosome 6, are the most abundantly expressed glutathione S-transferases in liver. In addition to metabolizing bilirubin and certain anti-cancer drugs in the liver, the alpha class of these enzymes exhibit glutathione peroxidase activity thereby protecting the cells from reactive oxygen species and the products of peroxidation.
