Sheep Glutamate Receptor, Ionotropic, N-Methyl-D-Aspartate 2C ELISA Kit
Principle of the Assay: GRIN2C ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GRIN2C antibody and an GRIN2C-HRP conjugate. The assay sample and buffer are incubated together with GRIN2C-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRIN2C concentration since GRIN2C from samples and GRIN2C-HRP conjugate compete for the anti-GRIN2C antibody binding site. Since the number of sites is limited, as more sites are occupied by GRIN2C from the sample, fewer sites are left to bind GRIN2C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRIN2C concentration in each sample is interpolated from this standard curve.

This gene encodes a subunit of the N-methyl-D-aspartate (NMDA) receptor, which is a subtype of ionotropic glutamate receptor. NMDA receptors are found in the central nervous system, are permeable to cations and have an important role in physiological processes such as learning, memory, and synaptic development. The receptor is a tetramer of different subunits (typically heterodimer of subunit 1 with one or more of subunits 2A-D), forming a channel that is permeable to calcium, potassium, and sodium, and whose properties are determined by subunit composition. Alterations in the subunit composition of the receptor are associated with pathophysiological conditions such as Parkinson's disease, Alzheimer's disease, depression, and schizophrenia.
