Biological Significance
Glutaredoxin(GRX), also known as thioltransferase, is a member of the thiol-disulfide oxidoreductase family. Glutraredoxin catalyzes the reversible reduction of protein-glutathionyl mixed disulfides to free sulfhydryl groups though a monothiol mechanism. Glutaredoxin-3 (Grx3/PICOT) is an essential protein involved in the regulation of signal transduction, for instance during immune cell activation and development of cardiac hypertrophy, presumably in response to redox signals. And it has been shown to interact with PRKCQ. Recombinant human Glutaredoxin-3, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.

NCBI Description

This gene encodes a member of the glutaredoxin family. Glutaredoxins are oxidoreductase enzymes that reduce a variety of substrates using glutathione as a cofactor. The encoded protein binds to and modulates the function of protein kinase C theta. The encoded protein may also inhibit apoptosis and play a role in cellular growth, and the expression of this gene may be a marker for cancer. Pseudogenes of this gene are located on the short arm of chromosomes 6 and 9. Alternatively spliced transcript variants have been observed for this gene. [provided by RefSeq, Dec 2010]

Uniprot Description

GLRX3: Critical negative regulator of cardiac hypertrophy and a positive inotropic regulator. May play a role in regulating the function of the thioredoxin system. Does not posses any thyoredoxin activity since it lacks the conserved motif that is essential for catalytic activity.

Protein type: Oxidoreductase

Chromosomal Location of Human Ortholog: 10q26

Cellular Component: cell cortex; Z disc

Molecular Function: protein binding; protein kinase C binding; electron carrier activity; metal ion binding; protein disulfide oxidoreductase activity; iron-sulfur cluster binding

Biological Process: cell redox homeostasis; regulation of the force of heart contraction

Product Notes

The GLRX3 glrx3 (Catalog #AAA836807) is a Recombinant Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Glutaredoxin3 can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Researchers should empirically determine the suitability of the GLRX3 glrx3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: 1-335 amino acids: MGSSHHHHHH SSGLVPRGSH MAAGAAEAAV AAVEEVGSAG QFEELLRLKA KSLLVVHFWA PWAPQCAQMN EVMAELAKEL PQVSFVKLEA EGVPEVSEKY EISSVPTFLF FKNSQKIDRL DGAHAPELTK KVQRHASSGS FLPSANEHLK EDLNLRLKKL THAAPCMLFM KGTPQEPRCG FSKQMVEILH KHNIQFSSFD IFSDEEVRQG LKAYSSWPTY PQLYVSGELI GGLDIIKELE ASEELDTICP KAPKLEERLK VLTNKASVML FMKGNKQEAK CGFSKQILEI LNSTGVEYET FDILEDEEVR QGLKAYSNWP TYPQLYVKGE LVGGLDIVKE LKENGELLPI LRGEN. It is sometimes possible for the material contained within the vial of "Glutaredoxin3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
