This assay has high sensitivity and excellent specificity for detection of GGT1. No significant cross-reactivity or interference between GGT1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GGT1 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This GGT1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken GGT1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: GGT1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GGT1 antibody and an GGT1-HRP conjugate. The assay sample and buffer are incubated together with GGT1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GGT1 concentration since GGT1 from samples and GGT1-HRP conjugate compete for the anti-GGT1 antibody binding site. Since the number of sites is limited, as more sites are occupied by GGT1 from the sample, fewer sites are left to bind GGT1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GGT1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The enzyme encoded by this gene catalyzes the transfer of the glutamyl moiety of glutathione to a variety of amino acids and dipeptide acceptors. The enzyme is composed of a heavy chain and a light chain, which are derived from a single precursor protein, and is present in tissues involved in absorption and secretion. This enzyme is a member of the gamma-glutamyltransferase protein family, of which many members have not yet been fully characterized and some of which may represent pseudogenes. This gene is classified as type I gamma-glutamyltransferase. Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Oct 2008]

GGT1: Initiates extracellular glutathione (GSH) breakdown, provides cells with a local cysteine supply and contributes to maintain intracellular GSH level. It is part of the cell antioxidant defense mechanism. Catalyzes the transfer of the glutamyl moiety of glutathione to amino acids and dipeptide acceptors. Alternatively, glutathione can be hydrolyzed to give Cys-Gly and gamma glutamate. Isoform 3 seems to be inactive. Defects in GGT1 are a cause of glutathionuria (GLUTH); also known as gamma-glutamyltranspeptidase deficiency. It is an autosomal recessive disease. Belongs to the gamma-glutamyltransferase family. 3 isoforms of the human protein are produced by alternative promoter.
