Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Intended Uses: The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of progesterone in serum, plasma and other biological fluids.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to progesterone has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled progesterone and unlabeled progesterone (Standards or samples) with the pre-coated antibody specific to progesterone. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of progesterone in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of progesterone in the sample.

NCBI Description

This gene encodes a member of the steroid receptor superfamily. The encoded protein mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy. This gene uses two distinct promotors and translation start sites in the first exon to produce several transcript variants, both protein coding and non-protein coding. Two of the isoforms (A and B) are identical except for an additional 165 amino acids found in the N-terminus of isoform B and mediate their own response genes and physiologic effects with little overlap. The steroid hormones and their receptors are involved in the regulation of eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. Depending on the isoform, progesterone receptor functions as transcriptional activator or repressor.
