GCA, also known as Grancalcin, is calcium-binding protein that is especially abundant in human neutrophils. It belongs to the penta EF-hand (PEF) subfamily of EF-hand proteins, which also comprises calpain, sorcin, peflin, and ALG-2. This protein undergoes important conformational changes upon binding of calcium, which subsequently exposes hydrophobic amino acid residues, which direct the protein to hydrophobic surfaces. It interacts with L-plastin, a protein known to have actin bundling activity, which suggests that GCA may play a role in the regulation of neutrophils adhesion. Recombinant human GCA protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography.

This gene encodes a calcium-binding protein that is abundant in neutrophils and macrophages. In the absence of divalent cation, this protein localizes to the cytosolic fraction; with magnesium alone, it partitions with the granule fraction; and in the presence of magnesium and calcium, it associates with both the granule and membrane fractions. Alternative splicing and use of alternative promoters results in multiple transcript variants. [provided by RefSeq, Aug 2016]
