Principle of the Assay: GAD65Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing Glutamic Acid Decarboxylase 65 antigen and an GAD65Ab-HRP conjugate. The assay sample and buffer are incubated together with GAD65Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAD65Ab concentration since GAD65Ab from samples and GAD65Ab-HRP conjugate compete for the Glutamic Acid Decarboxylase 65 antigen binding site. Since the number of sites is limited, as more sites are occupied by GAD65Ab from the sample, fewer sites are left to bind GAD65Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAD65Ab concentration in each sample is interpolated from this standard curve.

This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantigen and an autoreactive T cell target in insulin-dependent diabetes. This gene may also play a role in the stiff man syndrome. Deficiency in this enzyme has been shown to lead to pyridoxine dependency with seizures. Alternative splicing of this gene results in two products, the predominant 67-kD form and a less-frequent 25-kD form. [provided by RefSeq, Jul 2008]
