Principle of the Assay: FMOD ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FMOD antibody and an FMOD-HRP conjugate. The assay sample and buffer are incubated together with FMOD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FMOD concentration since FMOD from samples and FMOD-HRP conjugate compete for the anti-FMOD antibody binding site. Since the number of sites is limited, as more sites are occupied by FMOD from the sample, fewer sites are left to bind FMOD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FMOD concentration in each sample is interpolated from this standard curve.

NCBI Description

Fibromodulin belongs to the family of small interstitial proteoglycans. The encoded protein possesses a central region containing leucine-rich repeats with 4 keratan sulfate chains, flanked by terminal domains containing disulphide bonds. Owing to the interaction with type I and type II collagen fibrils and in vitro inhibition of fibrillogenesis, the encoded protein may play a role in the assembly of extracellular matrix. It may also regulate TGF-beta activities by sequestering TGF-beta into the extracellular matrix. Sequence variations in this gene may be associated with the pathogenesis of high myopia.
