Scientific Background: FADD or Fas-Associated protein with Death Domain is an adaptor molecule that mediates death signaling by the Fas-receptor, tumor necrosis factor receptor and TRAIL-receptor. FADD binds to these receptors via the C-terminus Death Domain which then unmasks the N-terminal effector domain of FADD thereby allowing it to recruit caspase-8 and activate the cysteine protease cascade leading to apoptosis (1). Cells lacking FADD are defective in intracellular double-stranded RNA (dsRNA)-activated gene expression, including production of type I (alpha/beta) interferons and are thus very susceptible to viral infection (2).

NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is an adaptor molecule that interacts with various cell surface receptors and mediates cell apoptotic signals. Through its C-terminal death domain, this protein can be recruited by TNFRSF6/Fas-receptor, tumor necrosis factor receptor, TNFRSF25, and TNFSF10/TRAIL-receptor, and thus it participates in the death signaling initiated by these receptors. Interaction of this protein with the receptors unmasks the N-terminal effector domain of this protein, which allows it to recruit caspase-8, and thereby activate the cysteine protease cascade. Knockout studies in mice also suggest the importance of this protein in early T cell development. [provided by RefSeq, Jul 2008]
Uniprot Description
Function: Apoptotic adaptor molecule that recruits caspase-8 or caspase-10 to the activated Fas (CD95) or TNFR-1 receptors. The resulting aggregate called the death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation. Active caspase-8 initiates the subsequent cascade of caspases mediating apoptosis. Involved in interferon-mediated antiviral immune response, playing a role in the positive regulation of interferon signaling. Ref.17 Ref.23 Ref.24 Ref.25
Subunit structure: Can self-associate. Interacts with CFLAR, PEA15 and MBD4. When phosphorylated, part of a complex containing HIPK3 and FAS. May interact with MAVS/IPS1. Interacts with MOCV v-CFLAR protein and LRDD. Interacts (via death domain) with FAS (via death domain). Interacts with CASP8. Ref.10 Ref.11 Ref.12 Ref.13 Ref.14 Ref.17 Ref.23 Ref.24 Ref.25
Tissue specificity: Expressed in a wide variety of tissues, except for peripheral blood mononuclear leukocytes.
Domain: Contains a death domain involved in the binding of the corresponding domain within Fas receptor. Ref.24The interaction between the FAS and FADD death domains is crucial for the formation of the death-inducing signaling complex (DISC). Ref.24
Involvement in disease: Infections, recurrent, associated with encephalopathy, hepatic dysfunction and cardiovascular malformations (IEHDCM) [MIM:613759]: A condition with biological features of autoimmune lymphoproliferative syndrome such as high-circulating CD4(-)CD8(-)TCR-alpha-beta(+) T-cell counts, and elevated IL10 and FASL levels. Affected individuals suffer from recurrent, stereotypical episodes of fever, encephalopathy, and mild liver dysfunction sometimes accompanied by generalized seizures. The episodes can be triggered by varicella zoster virus (VZV), measles mumps rubella (MMR) attenuated vaccine, parainfluenza virus, and Epstein-Barr virus (EBV).Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.17
Sequence similarities: Contains 1 death domain.Contains 1 DED (death effector) domain.

Product Notes
The FADD fadd (Catalog #AAA515129) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is His tag!!Expression System||E.coli!!Source Note||Recombinant full-length human FADD was expressed in E. coli cell. AAA Biotech's FADD can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the FADD fadd for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "FADD, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
