PRINCIPLE OF THE ASSAY EPOR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-EPOR antibody and an EPOR-HRP conjugate. The assay sample and buffer are incubated together with EPOR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensi ty of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the EPOR concentration since EPOR from samples and EPOR-HRP conjugate compete for the anti-EPOR antibody binding site. Since the number of sites is limited, as more sites are occupied by EPOR from the sample, fewer sites are left to bind EPOR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The EPOR concentration in each sample is interpolated from this standard curve.

This gene encodes the erythropoietin receptor which is a member of the cytokine receptor family. Upon erythropoietin binding, this receptor activates Jak2 tyrosine kinase which activates different intracellular pathways including: Ras/MAP kinase, phosphatidylinositol 3-kinase and STAT transcription factors. The stimulated erythropoietin receptor appears to have a role in erythroid cell survival. Defects in the erythropoietin receptor may produce erythroleukemia and familial erythrocytosis. Upon EPO stimulation, EPOR dimerizes triggering the JAK2/STAT5 signaling cascade. In some cell types, can also activate STAT1 and STAT3. Forms homodimers on EPO stimulation. Tyrosine-phosphorylated EpoR may bind several SH2 domain-containing proteins including LYN, the adapter protein APS, SHP-1, SHP-2, JAK2, PI3 kinases, STAT5A/B, SOCS3, and CRKL. Defects in the erythropoietin receptor may produce erythroleukemia and familial erythrocytosis. Three alternatively-spliced isoforms have been described. Isoform EPOR-T, missing the cytoplasmic tail, acts as a dominant-negative receptor of EPOR-mediated signaling.
