INTENDED USE This ECM1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken ECM1. This ELISA kit for research use only!
PRINCIPLE OF THE ASSAY ECM1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ECM1 antibody and an ECM1-HRP conjugate. The assay sample and buffer are incubated together with ECM1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ECM1 concentration since ECM1 from samples and ECM1-HRP conjugate compete for the anti-ECM1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ECM1 from the sample, fewer sites are left to bind ECM1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ECM1 concentration in each sample is interpolated from this standard curve.

Uniprot Description
ECM1: Involved in endochondral bone formation as negative regulator of bone mineralization. Stimulates the proliferation of endothelial cells and promotes angiogenesis. Inhibits MMP9 proteolytic activity. Defects in ECM1 are the cause of lipoid proteinosis (LiP); also known as lipoid proteinosis of Urbach and Wiethe or hyalinosis cutis et mucosae. LiP is a rare autosomal recessive disorder characterized by generalized thickening of skin, mucosae and certain viscera. Classical features include beaded eyelid papules and laryngeal infiltration leading to hoarseness. Histologically, there is widespread deposition of hyaline material and disruption/reduplication of basement membrane. 4 isoforms of the human protein are produced by alternative splicing.

Product Notes
The Chicken ECM1 (Catalog #AAA740791) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA740791 ELISA Kit recognizes Chicken ECM1. It is sometimes possible for the material contained within the vial of "Extracellular Matrix Protein 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
