Biological Activity
Assay#1: Determined by the dose-dependent stimulation of MIP-2 production by mouse spleen cells using a concentration range of 10-100 ng/ml. Assay#2: Measured by its ability to induce apoptosis in LN-18 cells (human glioblastoma cells). The expected ED50 for this effect is 40.0-60.0 ng/ml.
Related Product Information for TRAIL active protein
TRAIL is a cytotoxic protein, which activates rapid apoptosis in tumor cells, but not in normal cells. TRAIL induced apoptosis is achieved through binding to two death-signaling receptors, DR4 and DR5. These receptors belong to the TNFR superfamily of transmembrane proteins and contain a cytoplasmic "death domain", which activates the cell's apoptotic machinery. Recombinant murine TRAIL is a 174 amino acid polypeptide (20.0 kDa), consisting of the TNF homologous portion of the extracellular domain of the full length TRAIL protein.

Uniprot Description

Function: Cytokine that binds to TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and possibly also to TNFRSF11B/OPG. Induces apoptosis. Its activity may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and TNFRSF11B/OPG that cannot induce apoptosis.

Subunit structure: Homotrimer

By similarity.

Subcellular location: Membrane; Single-pass type II membrane protein

Potential.

Tissue specificity: Widespread.

Sequence similarities: Belongs to the tumor necrosis factor family.

Product Notes

The TRAIL tnfsf10 (Catalog #AAA691694) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The Mouse TRAIL reacts with Mouse and may cross-react with other species as described in the data sheet. The amino acid sequence is listed below: MRGGRPQKVA AHITGITRRS NSALIPISKD GKTLGQKIES WESSRK GHSF LNHVLFRNGE LVIEQEGLYY IYSQTYFRFQ EAEDASKMVS K DKVRTKQLV QYIYKYTSYP DPIVLMKSAR NSCWSRDAEY GLYSIYQ GGL FELKKNDRIF VSVTNEHLMD LDQEASFFGA FLIN. It is sometimes possible for the material contained within the vial of "TRAIL, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
