Reconstitution
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/m. Do not votex. This solution can be stored at 2 degree C to 8 degree C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20 degree C to -80 degree C.
Preparation and Storage
The lyophilized protein is stable at room temperature for 1 month and at 4 degree C for 6 months. Reconstituted working aliquots are stable for 1 week at 2 degree C to 8 degree C and for 12 months at -20 degree C to -80 degree C.
Avoid freeze/thaw cycles.
Related Product Information for TNF-alpha active protein
TNF-alpha is a pleiotropic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF family of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, auto immune diseases, rheumatoid arthritis, inflammation, and diabetes. Recombinant murine TNF-alpha is a soluble 156 amino acid protein (17.3 kDa) which corresponds to C-terminal extracellular domain of the full length transmembrane protein.

NCBI Description

This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. Members of this family are classified based on primary sequence, function, and structure. This protein is synthesized as a type-II transmembrane protein and is reported to be cleaved into products that exert distinct biological functions. It plays an important role in the innate immune response as well as regulating homeostasis but is also implicated in diseases of chronic inflammation. In mouse deficiency of this gene is associated with defects in response to bacterial infection, with defects in forming organized follicular dendritic cell networks and germinal centers, and with a lack of primary B cell follicles. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2013]

Uniprot Description

Function: Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells

By similarity.

Subunit structure: Interacts with SPPL2B

By similarity. Homotrimer.

Subcellular location: Cell membrane; Single-pass type II membrane protein. Tumor necrosis factor, membrane form: Membrane; Single-pass type II membrane protein

By similarity. Tumor necrosis factor, soluble form: Secreted. C-domain 1: Secreted

By similarity. C-domain 2: Secreted

By similarity.

Post-translational modification: The membrane-bound form is further proteolytically processed by SPPL2A or SPPL2B through regulated intramembrane proteolysis producing TNF intracellular domains (ICD1 and ICD2) released in the cytosol and TNF C-domain 1 and C-domain 2 secreted into the extracellular space

By similarity. The soluble form derives from the membrane form by proteolytic processing. The membrane form, but not the soluble form, is phosphorylated on serine residues. Dephosphorylation of the membrane form occurs by binding to soluble TNFRSF1A/TNFR1

By similarity.O-glycosylated; glycans contain galactose, N-acetylgalactosamine and N-acetylneuraminic acid

By similarity.

Sequence similarities: Belongs to the tumor necrosis factor family.

Product Notes

The TNF-alpha tnf (Catalog #AAA691749) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The Mouse TNF-alpha reacts with Mouse and may cross-react with other species as described in the data sheet. The amino acid sequence is listed below: MLRSSSQNSS DKPVAHVVAN HQVEEQLEWL SQRANALLAN GMDLKD NQLV VPADGLYLVY SQVLFKGQGC PDYVLLTHTV SRFAISYQEK V NLLSAVKSP CPKDTPEGAE LKPWYEPIYL GGVFQLEKGD QLSAEVN LPK YLDFAESGQV YFGVIAL. It is sometimes possible for the material contained within the vial of "TNF-alpha, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
