Stability
The lyophilized protein is stable at room temperature for 1 month and at 4°C for 6 months. Reconstituted working aliquots are stable for 1 week at 2°C to 8°C and for 3 months at -20°C to -80°C.
Reconstitution
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/m. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containinga carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C

Tested by its ability to down-regulate lipid biosynthesis in aortic smooth muscle cells. The effective concentration was found to be 4 uM.

Human Apo-SAA is a 104 amino acid polypeptide that circulates primarily in association with high-density lipoproteins (HDL). The level of Apo-SAA, normally 1-5 ug/ml in plasma, increases 500-1000 fold within 24 hours of an inflammatory stimulus and, under these conditions, is the most abundant HDL Apolipoprotein. Thehuman SAA gene codes for a 122 amino acid polypeptide, which contains an 18 amino acid N-terminal signal sequence. Recombinant Apo-SAA is a consensus SAA molecule corresponding to human Apo-SAA1alpha except for the presence of an N-terminal methionine andsubstitution of asparagine for aspartic acid at position 60 and arginine for histidine at position 71(the last two substituted residues are present in apo-SAA2beta).The calculated molecular weight of recombinant human Apo-SAAis 11.7 kDa.

NCBI Description

This gene encodes a member of the serum amyloid A family of apolipoproteins. The encoded protein is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. This protein also plays an important role in HDL metabolism and cholesterol homeostasis. High levels of this protein are associated with chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, Alzheimer's disease and Crohn's disease. This protein may also be a potential biomarker for certain tumors. Alternate splicing results in multiple transcript variants that encode the same protein. A pseudogene of this gene is found on chromosome 11.[provided by RefSeq, Jun 2012]

Subcellular location: Secreted.

Tissue specificity: Expressed by the liver; secreted in plasma. Ref.18

Induction: Upon cytokine stimulation.

Post-translational modification: This protein is the precursor of amyloid protein A, which is formed by the removal of approximately 24 residues from the C-terminal end.

Polymorphism: At least 5 different SAA1 alleles have been described: SAA1.1 (SAA1alpha), SAA1.2 (SAA1beta), SAA1.3 (SAA1gamma), SAA1.4 (SAA1delta), SAA1.5 (also named SAA1beta but which differs from SAA1.2). We use here the revised nomenclature described in Ref.21. The sequence shown is that of SAA1.1.

Involvement in disease: Reactive, secondary amyloidosis is characterized by the extracellular accumulation in various tissues of the SAA1 protein. These deposits are highly insoluble and resistant to proteolysis; they disrupt tissue structure and compromise function. Ref.14Elevated serum SAA1 protein levels may be associated with lung cancer. Ref.14
