1. UV spectroscopy at 280 nm using the absorbency value of 1.26 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of TNF-alpa as a Reference Standard.
Stability: Lyophilized Porcine TNF-a although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution TNF-alpha should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.
Recombinant Porcine TNF-a produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 157 amino acids and having a molecular mass of 17,274 Dalton. Porcine TNF-alpha is purified by standard chromatographic techniques.
NCBI and Uniprot Product Information
Uniprot Description
Function: Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells
By similarity.
Subunit structure: Homotrimer. Interacts with SPPL2B
By similarity.
Subcellular location: Cell membrane; Single-pass type II membrane protein
By similarity. Tumor necrosis factor, membrane form: Membrane; Single-pass type II membrane protein
By similarity. Tumor necrosis factor, soluble form: Secreted
By similarity. C-domain 1: Secreted
By similarity. C-domain 2: Secreted
By similarity.
Post-translational modification: The soluble form derives from the membrane form by proteolytic processing. The membrane-bound form is further proteolytically processed by SPPL2A or SPPL2B through regulated intramembrane proteolysis producing TNF intracellular domains (ICD1 and ICD2) released in the cytosol and TNF C-domain 1 and C-domain 2 secreted into the extracellular space
By similarity. The membrane form, but not the soluble form, is phosphorylated on serine residues. Dephosphorylation of the membrane form occurs by binding to soluble TNFRSF1A/TNFR1
By similarity.O-glycosylated; glycans contain galactose, N-acetylgalactosamine and N-acetylneuraminic acid
By similarity.
Sequence similarities: Belongs to the tumor necrosis factor family.

Product Notes
The TNF tnf (Catalog #AAA650765) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Leu-Ar g-Ser-Ser. It is sometimes possible for the material contained within the vial of "Tumor Necrosis Factor alpha, Porcine, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
