95% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Synonyms
MCAF; Recombinant; Human (MCP-1; Macrophage/Monocyte Chemotactic and Activating Factor); MCAF active protein
Ordering
For Research Use Only!
Host
E Coli
Purity/Purification
Highly Purified
95% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Form/Format
Supplied as a lyophilized powder. No additives.
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Pro-Asp-Ala-Ile.
Biological Activity
MCP-1 is fully biologically active when compared to standard. The specific activity as determined by the ability of MCP-1 to chemoattract human Monocytes using a concentration of 5-20ng/ml.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of rHuMCP-1.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm.
2. Analysis by RP-HPLC, using a standard solution of MCP-1 as a Reference Standard.
Reconstitution
Reconstitute the lyophilized rHuMCP-1 in sterile 18M -cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI and Uniprot Product Information

NCBI GI #
NCBI Official Full Name
MBD1-containing chromatin associated factor

NCBI Description

ATF7IP is a multifunctional nuclear protein that associates with heterochromatin. It can act as a transcriptional coactivator or corepressor depending upon its binding partners (summary by Liu et al., 2009 [PubMed 19106100]).[supplied by OMIM, Nov 2010]

Uniprot Description

ATF7IP: Recruiter that couples transcriptional factors to general transcription apparatus and thereby modulates transcription regulation and chromatin formation. Can both act as an activator or a repressor depending on the context. Mediates MBD1-dependent transcriptional repression, probably by recruiting complexes containing SETDB1. Required to stimulate histone methyltransferase activity of SETDB1 and facilitate the conversion of dimethylated to trimethylated H3 'Lys-9' (H3K9me3). The complex formed with MBD1 and SETDB1 represses transcription and couples DNA methylation and histone H3 'Lys-9' trimethylation (H3K9me3). Facilitates telomerase TERT and TERC gene expression by SP1 in cancer cells. Belongs to the MCAF family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription, coactivator/corepressor

Chromosomal Location of Human Ortholog: 12p13.1

Cellular Component: nucleoplasm; transcription factor complex; cytoplasm; nucleus

Molecular Function: protein binding; ATPase activity; transcription corepressor activity

Biological Process: establishment and/or maintenance of chromatin architecture; transcription, DNA-dependent; viral reproduction; positive regulation of transcription, DNA-dependent; DNA methylation; negative regulation of transcription from RNA polymerase II promoter; negative regulation of transcription, DNA-dependent; regulation of transcriptional preinitiation complex assembly

Product Notes

The MCAF (Catalog #AAA650850) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Pro-As p-Ala-Ile. It is sometimes possible for the material contained within the vial of "MCAF, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
