Lyophilized MIP-1alpha is soluble in water and most aqueous buffers and can be reconstituted in water to a concentration of 0.1mg/ml. This solution can be diluted into other buffered solutions or stored frozen for future use.

Both MIP-1 alpha and MIP-1 beta are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1 alpha and MIP-1 beta exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1 alpha selectively attracting CD8+ lymphocytes and MIP-1 beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1 alpha and MIP-1 beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1 alpha and MIP-1 beta are active on human and murine hematopoietic cells.

Recombinant human MIP-1 alpha is a 7.8kD protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines.

NCBI Description

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.

Uniprot Description

Function: Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Ref.8

Subunit structure: Self-associates. Also heterodimer of MIP-1-alpha(4-69) and MIP-1-beta(3-69). Ref.11

Subcellular location: Secreted.

Induction: By TPA or PHA (TPA = 12-O-tetradecanoyl phorbol-13 acetate (tumor promoter); PHA = phytohemagglutinin (T-cell mitogen)).

Post-translational modification: N-terminal processed form LD78-alpha(4-69) is produced by proteolytic cleavage after secretion from HTLV1-transformed T-cells.

Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.
