Sequence
The sequence of the N-terminal amino acids could not be determined indicating that the PEG moiety is attached to the N-terminus.
Solubility
It is recommended to reconstitute the lyophilized pegylated mouse IL22 Y51A in sterile water or sterile 0.4% NaHCO3 adjusted to pH 8-9, not less than 100ug/ml, which can then be further diluted with other aqueous solutions.
Protein Content
Protein quantitation of mouse interleukin 22 Y51A mutant was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.18 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the DNAman computer analysis program of protein sequences.

The purified mouse pegylated IL22 consists of > 90% monomers as determined by gel-filtration chromatography.

Less than 0.05 ng/ug (0.5 EU/ug) of recombinant pegylated IL-22 Y51A.
Formulation
The protein was lyophilized from a concentrated (1.0 mg/ml) solution with 0.003mM NaHCO3.
Biological Activity
murine IL-22 Y51A mutant is biologically active, capable of full inhibition of mouse IL-22 induced STAT3 phosphorylation in HepG cells. Its inhibitory acrtivity in vitro is ~ 10-20% compared to the non-pegylated mIL22 (Y51A) mutant.
Preparation and Storage
Lyophilized pegylated mouse IL22 although stable at room temperature for several days, should be stored desiccated below -20 degree C. Upon reconstitution at > 0.1 pegylated mIL22 mg/ml and up to 2 mg/ml and filter sterilization it can be stored at 4 degree C or even room temperature for several weeks making it suitable for long term infusion studies using osmotic pumps. At lower concentration addition of a carrier protein (0.1% HSA or BSA) is suggested. Please prevent freeze-thaw cycles.
