Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of DUB. No significant cross-reactivity or interference between DUB and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DUB and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL

Related Product Information for DUB elisa kit
Intended Uses: This DUB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human DUB. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DUB ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DUB antibody and an DUB-HRP conjugate. The assay sample and buffer are incubated together with DUB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DUB concentration since DUB from samples and DUB-HRP conjugate compete for the anti-DUB antibody binding site. Since the number of sites is limited, as more sites are occupied by DUB from the sample, fewer sites are left to bind DUB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DUB concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the ubiquitin specific protease (USP) family of deubiquitinating enzymes. USP enzymes play critical roles in ubiquitin-dependent processes through polyubiquitin chain disassembly and hydrolysis of ubiquitin-substrate bonds. The encoded protein associates with the COP9 signalosome, and also plays a role in transforming growth factor beta signalling through deubiquitination of receptor-activated SMAD transcription factors. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene, and a pseudogene of this gene is located on the long arm of chromosome 2. Uniprot Description

USP15: Protease that degrades 'Lys-48'-linked polyubiquitin chains. Protects target proteins against proteasomal degradation. Protects APC and human papillomavirus type 16 protein E6 against degradation via the ubiquitin proteasome pathway. Belongs to the peptidase C19 family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human DUB usp15 (Catalog #AAA727833) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA727833 ELISA Kit recognizes Human DUB. It is sometimes possible for the material contained within the vial of "Deubiquitinating enzyme, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
