Intended Uses: This DRD4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine DRD4. This ELISA kit for research use only!

Principle of the Assay: DRD4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DRD4 antibody and an DRD4-HRP conjugate. The assay sample and buffer are incubated together with DRD4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DRD4 concentration since DRD4 from samples and DRD4-HRP conjugate compete for the anti-DRD4 antibody binding site. Since the number of sites is limited, as more sites are occupied by DRD4 from the sample, fewer sites are left to bind DRD4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DRD4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the D4 subtype of the dopamine receptor. The D4 subtype is a G-protein coupled receptor which inhibits adenylyl cyclase. It is a target for drugs which treat schizophrenia and Parkinson disease. Mutations in this gene have been associated with various behavioral phenotypes, including autonomic nervous system dysfunction, attention deficit/hyperactivity disorder, and the personality trait of novelty seeking. This gene contains a polymorphic number (2-10 copies) of tandem 48 nt repeats; the sequence shown contains four repeats. [provided by RefSeq, Jul 2008]
