Dihydrolipoamide succinyltransferase, which is a component of the structural core of the alpha-keto glutarate dehydrogenase complex, was studied by Nakano et al. (1993), who isolated a cDNA from a human fibroblast cDNA library. Amino acid sequence analysis supported their previous observation (Nakano et al., 1993) that human dihydrolipoamide succinyltransferase lacks a sequence motif for an E1 and/or E3 binding site. By Northern blot analysis, Ali et al. (1994) detected ubiquitous expression of DLST in peripheral tissues and brain.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate DLST in samples. An antibody specific for DLST has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDLST present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DLST is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DLST bound in the initial step. The color development is stopped and the intensity of the color is measured.
