Canine Type II iodothyronine deiodinase ELISA Kit
Principle of the Assay: DIO2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DIO2 antibody and an DIO2-HRP conjugate. The assay sample and buffer are incubated together with DIO2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DIO2 concentration since DIO2 from samples and DIO2-HRP conjugate compete for the anti-DIO2 antibody binding site. Since the number of sites is limited, as more sites are occupied by DIO2 from the sample, fewer sites are left to bind DIO2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DIO2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene belongs to the iodothyronine deiodinase family. It activates thyroid hormone by converting the prohormone thyroxine (T4) by outer ring deiodination (ORD) to bioactive 3,3',5-triiodothyronine (T3). It is highly expressed in the thyroid, and may contribute significantly to the relative increase in thyroidal T3 production in patients with Graves disease and thyroid adenomas. This protein contains selenocysteine (Sec) residues encoded by the UGA codon, which normally signals translation termination. The 3' UTR of Sec-containing genes have a common stem-loop structure, the sec insertion sequence (SECIS), which is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]
