Intended Uses: This CXCR2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of MouseCXCR2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CXCR2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CXCR2 antibody and an CXCR2-HRP conjugate. The assay sample and buffer are incubated together with CXCR2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CXCR2 concentration since CXCR2 from samples and CXCR2-HRP conjugate compete for the anti-CXCR2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CXCR2 from the sample, fewer sites are left to bind CXCR2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CXCR2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the G-protein-coupled receptor family. This protein is a receptor for interleukin 8 (IL8). It binds to IL8 with high affinity, and transduces the signal through a G-protein activated second messenger system. This receptor also binds to chemokine (C-X-C motif) ligand 1 (CXCL1/MGSA), a protein with melanoma growth stimulating activity, and has been shown to be a major component required for serum-dependent melanoma cell growth. This receptor mediates neutrophil migration to sites of inflammation. The angiogenic effects of IL8 in intestinal microvascular endothelial cells are found to be mediated by this receptor. Knockout studies in mice suggested that this receptor controls the positioning of oligodendrocyte precursors in developing spinal cord by arresting their migration. This gene, IL8RA, a gene encoding another high affinity IL8 receptor, as well as IL8RBP, a pseudogene of IL8RB, form a gene cluster in a region mapped to chromosome 2q33-q36. Alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Nov 2009]
