Principle of the assay: human Cathepsin S ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Cathepsin S has been precoated onto 96-well plates. Standards(NSO, Q17-I331 & S109-I331) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Cathepsin S is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Cathepsin S amount of sample captured in plate.
Background: Cathepsin S, also known as CTSS, is a protein that in humans is encoded by the CTSS gene. A member of the peptidase C1 family, CTSS gene is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. By fluorescence in situ hybridization (FISH), this gene is mapped to 1q21.3. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. The bond-specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N.

NCBI Description

The protein encoded by this gene, a member of the peptidase C1 family, is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Dec 2010]

Uniprot Description

CTSS: Thiol protease. Key protease responsible for the removal of the invariant chain from MHC class II molecules. The bond- specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N. Belongs to the peptidase C1 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Protease; EC 3.4.22.27

Chromosomal Location of Human Ortholog: 1q21

Cellular Component: lysosomal lumen; extracellular space; intracellular membrane-bound organelle; membrane; lysosome; extracellular region

Product Notes

The Human CTSS ctss (Catalog #AAA177353) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA177353 ELISA Kit recognizes Human CTSS. It is sometimes possible for the material contained within the vial of "Cathepsin S, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
