Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of human Cys -C. No significant cross-reactivity or interference between human Cys -C and analogues was observed.
Sequence Length
146
Samples
Serum, plasma, urine, saliva, tissue homogenates
Assay Type
Sandwich
Detection Range
7.81 ng/ml -500 ng/ml.
Sensitivity
The minimum detectable dose of human Cys -C is typically less than 5.824 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.

Priciple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for Cys-C has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Cys-C present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Cys-C is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Cys-C bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions, where they appear to provide protective functions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes the most abundant extracellular inhibitor of cysteine proteases, which is found in high concentrations in biological fluids and is expressed in virtually all organs of the body. A mutation in this gene has been associated with amyloid angiopathy. Expression of this protein in vascular wall smooth muscle cells is severely reduced in both atherosclerotic and aneurysmal aortic lesions, establishing its role in vascular disease. Alternative splicing results in multiple transcript variants encoding a single protein. [provided by RefSeq, Dec 2013]

Uniprot Description

Function: As an inhibitor of cysteine proteinases, this protein is thought to serve an important physiological role as a local regulator of this enzyme activity.

Product Notes

The Human CST3 cst3 (Catalog #AAA700210) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA700210 ELISA Kit recognizes Human CST3. It is sometimes possible for the material contained within the vial of "cystatin C, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
