Principle of the Assay: CRH-R ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CRH-R antibody and an CRH-R-HRP conjugate. The assay sample and buffer are incubated together with CRH-R-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CRH-R concentration since CRH-R from samples and CRH-R-HRP conjugate compete for the anti-CRH-R antibody binding site. Since the number of sites is limited, as more sites are occupied by CRH-R from the sample, fewer sites are left to bind CRH-R-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CRH-R concentration in each sample is interpolated from this standard curve.

This gene encodes a G-protein coupled receptor that binds neuropeptides of the corticotropin releasing hormone family that are major regulators of the hypothalamic-pituitary-adrenal pathway. The encoded protein is essential for the activation of signal transduction pathways that regulate diverse physiological processes including stress, reproduction, immune response and obesity. Alternative splicing results in multiple transcript variants, one of which represents a read-through transcript with the neighboring gene MGC57346. [provided by RefSeq, Jan 2012]
