Applicable Applications for WNT9B active protein
SDS-PAGE
Application Notes
Recombinant WNT9B migrates as an approximately 37 kDa protein due to glycosylation under reducing conditions in SDS-PAGE.
Species
Human
Source Note
Optimized DNA sequence encoding Human WNT9B mature chain including a C-terminal 6xHis tag was expressed in CHO. Recombinant Human Protein Wnt-9b encodes amino acid residues 23-357.
Biological Activity
Determined by its ability to induce alkaline phosphatase production by CCL-226 cells.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/ug(1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this product can be stored in working aliquots at 2 degree C - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

NCBI Description

The WNT gene family consists of structurally related genes that encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. Study of its expression in the teratocarcinoma cell line NT2 suggests that it may be implicated in the early process of neuronal differentiation of NT2 cells induced by retinoic acid. This gene is clustered with WNT3, another family member, in the chromosome 17q21 region. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Feb 2016]

Uniprot Description

Ligand for members of the frizzled family of seven transmembrane receptors (Probable). Functions in the canonical Wnt/beta-catenin signaling pathway. Required for normal embryonic kidney development, and for normal development of the urogenital tract, including uterus and part of the oviduct and the upper vagina in females, and epididymis and vas deferens in males. Activates a signaling cascade in the metanephric mesenchyme that induces tubulogenesis. Acts upstream of WNT4 in the signaling pathways that mediate development of kidney tubules and the Muellerian ducts. Plays a role in cranofacial development and is required for normal fusion of the palate during embryonic development ().

Product Notes

The WNT9B wnt9b (Catalog #AAA553628) is an Active Protein produced from CHO and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's WNT9B can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Recombinant WNT9B migrates as an approximately 37 kDa protein due to glycosylation under reducing conditions in SDS-PAGE. Researchers should empirically determine the suitability of the WNT9B wnt9b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: SYFGLTGREV LTPFPGLGTA AAPAQGGAHL KQCDLLKLSR RQKQLCRREP GLAETLRDAA HLGLLECQFQ FRHERWNCSL EGRMGLLKRG FKETAFLYAV SSAALTHTLA RACSAGRMER CTCDDSPGLE SRQAWQWGVC GDNLKYSTKF LSNFLGSKRG NKDLRARADA HNTHVGIKAV KSGLRTTCKC HGVSGSCAVR TCWKQLSPFR ETGQVLKLRY DSAVKVSSAT NEALGRLELW APARQGSLTK GLAPRSGDLV YMEDSPSFCR PSKYSPGTAG RVCSREASCS SLCCGRGYDT QSRLVAFSCH CQVQWCCYVE CQQCVQEELV YTCKH . It is sometimes possible for the material contained within the vial of "WNT9B, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
