(Image: Western blots of homogenates of SH-SY5Y cells, a human neuroblastoma cell line. Lane A shows blotting with Neurofilament-M, which reveals a strong NF-M band at ~150kDa. Lanes B and C are homogenates of SH-SY5Y cells which were treated with maitotoxin to activate caspase family enzymes (ref. 2). Now a ~100kDa band is seen in addition o the major NF-M band. This corresponds to the C-terminal segment of NF-M which is an in vivo calpain degradation product of human NF-M. Protocol on datasheet.)

Neurofilaments can be defined as the intermediate or 10nm filaments found in specifically in neuronal cells. In the electron microscope neurofilaments appears as 10nm diameter fibres of indeterminate length which generally have fine wispy protrusions from their sides. They are found particularly abundantly in axons of large projection neurons. Antibodies to the various neurofilament subunits are very useful cell type markers since the proteins are among the most abundant of the nervous system, are expressed only in neurons, and are biochemically very stable.

Neurofilament-M (NF-M) is the medium or middle runs at about 145-160kDa.

NCBI Description

Neurofilaments are type IV intermediate filament heteropolymers composed of light, medium, and heavy chains. Neurofilaments comprise the axoskeleton and functionally maintain neuronal caliber. They may also play a role in intracellular transport to axons and dendrites. This gene encodes the medium neurofilament protein. This protein is commonly used as a biomarker of neuronal damage. Alternative splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Oct 2008]

Uniprot Description

NFM: a neurofilament intermediate filament protein. Neurofilaments usually contain three intermediate filament proteins: L, M, and H which are involved in the maintenance of neuronal caliber. The levels of phosphorylation are altered developmentally and coincident with a change in the neurofilament function.

Chromosomal Location of Human Ortholog: 8p21

Cellular Component: axon; neurofilament; neuromuscular junction

Molecular Function: protein binding; structural constituent of cytoskeleton; microtubule binding

Biological Process: neurofilament bundle assembly; regulation of axon diameter; axon cargo transport; microtubule cytoskeleton organization and biogenesis

Product Notes

The NEFM nefm (Catalog #AAA555399) is an Antibody and is intended for research purposes only. The product is available for immediate purchase. The Neurofilament NF-M reacts with Chicken, Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Neurofilament NF-M can be used in a range of immunoassay formats including, but not limited to, Immunocytochemistry (ICC), Immunohistochemistry (IHC), Western Blot (WB). Researchers should empirically determine the suitability of the NEFM nefm for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Neurofilament NF-M, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
