Description: This assay employs a two-site sandwich ELISA to quantitate DCLRE1C in samples. An antibody specific for DCLRE1C has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DCLRE1C present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DCLRE1C is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DCLRE1C bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: DCLRE1C encodes a nuclear protein that is involved in V(D)J recombination and DNA repair. The protein has single-strand-specific 5'-3' exonuclease activity; it also exhibits endonuclease activity on 5' and 3' overhangs and hairpins when complexed with protein kinase, DNA-activated, catalytic polypeptide.The purified Artemis protein alone possesses single-strand-specific 5-prime-to-3-prime exonuclease activity. Upon complex formation, PRKDC phosphorylates Artemis, and Artemis acquires endonucleolytic activity on 5-prime and 3-prime overhangs, as well as hairpins. PRKDC regulates Artemis by both phosphorylation and complex formation to permit enzymatic activities that are critical for the hairpin-opening step of V(D)J recombination and for the 5-prime and 3-prime overhang processing in nonhomologous DNA end joining.

Uniprot Description

Required for V(D)J recombination, the process by which exons encoding the antigen-binding domains of immunoglobulins and T-cell receptor proteins are assembled from individual V, (D), and J gene segments. V(D)J recombination is initiated by the lymphoid specific RAG endonuclease complex, which generates site specific DNA double strand breaks (DSBs). These DSBs present two types of DNA end structures: hairpin sealed coding ends and phosphorylated blunt signal ends. These ends are independently repaired by the non homologous end joining (NHEJ) pathway to form coding and signal joints respectively. This protein exhibits single-strand specific 5'-3' exonuclease activity in isolation, and acquires endonucleolytic activity on 5' and 3' hairpins and overhangs when in a complex with PRKDC. The latter activity is required specifically for the resolution of closed hairpins prior to the formation of the coding joint. May also be required for the repair of complex DSBs induced by ionizing radiation, which require substantial end-processing prior to religation by NHEJ ().
