Principle of the Assay: gamma Secretase ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-gamma Secretase antibody and an gamma Secretase-HRP conjugate. The assay sample and buffer are incubated together with gamma Secretase-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the gamma Secretase concentration since gamma Secretase from samples and gamma Secretase-HRP conjugate compete for the anti-gamma Secretase antibody binding site. Since the number of sites is limited, as more sites are occupied by gamma Secretase from the sample, fewer sites are left to bind gamma Secretase-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The gamma Secretase concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a glycogen branching enzyme that catalyzes the transfer of alpha-1,4-linked glucosyl units from the outer end of a glycogen chain to an alpha-1,6 position on the same or a neighboring glycogen chain. Branching of the chains is essential to increase the solubility of the glycogen molecule and, consequently, in reducing the osmotic pressure within cells. Highest level of this enzyme are found in liver and muscle. Mutations in this gene are associated with glycogen storage disease IV (also known as Andersen's disease). [provided by RefSeq, Jul 2008]
