This assay employs a two-site sandwich ELISA to quantitate DES in samples. An antibody specific for DES has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DES present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DES is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DES bound in the initial step. The color development is stopped and the intensity of the color is measured.

Muscle-specific type III intermediate filament essential for proper muscular structure and function. Plays a crucial role in maintaining the structure of sarcomeres, inter-connecting the Z-disks and forming the myofibrils, linking them not only to the sarcolemmal cytoskeleton, but also to the nucleus and mitochondria, thus providing strength for the muscle fiber during activity. In adult striated muscle they form a fibrous network connecting myofibrils to each other and to the plasma membrane from the periphery of the Z-line structures.
