Principle of the Assay: CENPH ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CENPH antibody and an CENPH-HRP conjugate. The assay sample and buffer are incubated together with CENPH-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CENPH concentration since CENPH from samples and CENPH-HRP conjugate compete for the anti-CENPH antibody binding site. Since the number of sites is limited, as more sites are occupied by CENPH from the sample, fewer sites are left to bind CENPH-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CENPH concentration in each sample is interpolated from this standard curve.

Centromere and kinetochore proteins play a critical role in centromere structure, kinetochore formation, and sister chromatid separation. The protein encoded by this gene colocalizes with inner kinetochore plate proteins CENP-A and CENP-C in both interphase and metaphase. It localizes outside of centromeric heterochromatin, where CENP-B is localized, and inside the kinetochore corona, where CENP-E is localized during prometaphase. It is thought that this protein can bind to itself, as well as to CENP-A, CENP-B or CENP-C. Multimers of the protein localize constitutively to the inner kinetochore plate and play an important role in the organization and function of the active centromere-kinetochore complex.
