Principle of the Assay||The Human CD66a ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CD66a in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human CD66a coated on a 96-well plate. Standards and samples are pipetted into the wells and CD66a present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD66a antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD66a bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: CEACAM1, also known as CD66a (Cluster of Differentiation 66a), is a human glycoprotein encoded by a gene. This gene encodes a member of the carcinoembryonic antigen (CEA) gene family, which belongs to the immunoglobulin superfamily. CEACAM1 is mapped to 19q13.2 in human. The encoded protein was originally described in bile ducts of liver as biliary glycoprotein. Subsequently, it was found to be a cell-cell adhesion molecule detected on leukocytes, epithelia, and endothelia. The encoded protein mediates cell adhesion via homophilic as well as heterophilic binding to other proteins of the subgroup. Multiple cellular activities have been attributed to the encoded protein, including roles in the differentiation and arrangement of tissue three-dimensional structure, angiogenesis, apoptosis, tumor suppression, metastasis, and the modulation of innate and adaptive immune responses.

NCBI Description

This gene encodes a member of the carcinoembryonic antigen (CEA) gene family, which belongs to the immunoglobulin superfamily. Two subgroups of the CEA family, the CEA cell adhesion molecules and the pregnancy-specific glycoproteins, are located within a 1.2 Mb cluster on the long arm of chromosome 19. Eleven pseudogenes of the CEA cell adhesion molecule subgroup are also found in the cluster. The encoded protein was originally described in bile ducts of liver as biliary glycoprotein. Subsequently, it was found to be a cell-cell adhesion molecule detected on leukocytes, epithelia, and endothelia. The encoded protein mediates cell adhesion via homophilic as well as heterophilic binding to other proteins of the subgroup. Multiple cellular activities have been attributed to the encoded protein, including roles in the differentiation and arrangement of tissue three-dimensional structure, angiogenesis, apoptosis, tumor suppression, metastasis, and the modulation of innate and adaptive immune responses. Belongs to the immunoglobulin superfamily and the CEA family. Abundantly expressed in epithelia, vessel endothelia, trophoblasts, platelets and neutrophilic granulocytes. Also present in many cell types of the immune system such as B cells, T cells, NK cells, macrophages and dendritic cells. Five alternatively spliced isoforms have been described. Two isoforms are type I membrane proteins, while three are secreted.
