Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cyclin-dependent kinase 12. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for Cyclin-dependent kinase 12 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain Cyclin-dependent kinase 12, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Cyclin-dependent kinase 12 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

CDK12: a Ser/Thr protein kinase of the CMGC group and CDK family. Phosphorylates the C-terminal heptapeptide repeat domain (CTD) of the largest RNA polymerase II subunit RPB1, thereby acting as a key regulator of transcription elongation. Required for RNA splicing, possibly by phosphorylating SRSF1/SF2. Interacts with CCNL1 and CCNL2. Colocalized with nuclear speckles throughout interphase. Has a highly phosphorylated arginine/serine-rich (RS) domain N-terminal to the kinase domain. Chromosomal aberrations involving CDK12 may be a cause gastric cancer. Deletions within 17q12 region producing fusion transcripts with ERBB2, leading to CDK12-ERBB2 fusion leading to trunctated CDK12 protein not in-frame with ERBB2. Three isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mouse Cdk12 cdk12 (Catalog #AAA288487) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA288487 ELISA Kit recognizes Mouse Cdk12. It is sometimes possible for the material contained within the vial of "Cyclin-dependent kinase 12, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
