Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for VE-cad has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any VE-cad present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VE-cad is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VE-cad bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene is a classical cadherin from the cadherin superfamily and is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. An alternative splice variant has been described but its full length sequence has not been determined. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. This cadherin may play a important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. It associates with alpha-catenin forming a link to the cytoskeleton. Acts in concert with KRIT1 to establish and maintain correct endothelial cell polarity and vascular lumen. These effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. Required for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction.2 PublicationsManual assertion based on experiment in:Ref.10

Product Notes

The Human CDH5 cdh5 (Catalog #AAA703368) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA703368 ELISA Kit recognizes Human CDH5. It is sometimes possible for the material contained within the vial of "cadherin 5, type 2 (vascular endothelium), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
