Principle of the Assay: CD74 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CD74 antibody and an CD74-HRP conjugate. The assay sample and buffer are incubated together with CD74-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD74 concentration since CD74 from samples and CD74-HRP conjugate compete for the anti-CD74 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD74 from the sample, fewer sites are left to bind CD74-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD74 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF) which, when bound to the encoded protein, initiates survival pathways and cell proliferation. This protein also interacts with amyloid precursor protein (APP) and suppresses the production of amyloid beta (Abeta). Multiple alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]

Serves as cell surface receptor for the cytokine MIF. A chromosomal aberration involving CD74 is found in a non-small cell lung tumor. Results in the formation of a CD74- ROS1 chimeric protein. 3 isoforms of the human protein are produced by alternative splicing.
